Cloning and Expression Analysis of Phytoene Desaturase (PDS) Gene from Mango (<i>Mangifera indica</i> L)

Phytoene desaturase (PDS) affects the synthesis of carotenoids. It is a key gene in carotenoids biosynthesis pathway. In order to study function PDS  gene from mango fruits, phytoene dehydrogenase ( ) ‘Guifei’ fruit was obtained with RACE methods. The full-length cDNA sequence 1 820 bp, open reading frame 650 encoding 549 amino acids, molecular weight 61.34 KD, and isoelectric point 6.78. cluster analysis found that protein had close relationship grapefruit, cantaloupe, papaya. Its acid composition mainly alanine (ALa), leucine (Leu), valine (Val), so on. Expression different varieties by PCR showed: high expression red green low volume. Prediction domain, tertiary structure, interaction its protein, contain phytoene-desat, PLNO2487 superfamily, other domains. use STRING database interacted proteins such as PSY, ZDS, CRTISO. This research will be valuable for understanding mechanism regulation carotenoid can served basis metabolic engineering mango.
